Top: The 4.7 kb and 2/7 kb HindIII fragments diagnostic of the wild-type (WT) and targeted Mob1a alleles, respectively, are shown. WT allele: A portion of the mouse Mob1a WT locus showing the KpnI (K) HindIII (H), and BamHI (B) sites. Targeting vector: Targeting vector with the hygromycin resistance gene (Hygr) in reverse orientation to Mob1a transcription. Exon 2 was flanked by two loxP sequences (arrowheads), with a third loxP sequence flanking Hygr. Targeted allele: The mutated Mob1ata allele containing three loxP sequences and the Hygr gene. Flox allele: Mob1atm1.1Asuz (Mob1aflox) allele produced after Cre-mediated Hygr deletion. Delta allele: Mob1atm1.2Asuz (Mob1adelta) allele produced after Cre-mediated deletion. Bottom: The 9.1 kb, 6.5 kb, and 4.7 kb KpnI fragments diagnostic of the WT Mob1a allele, the Mob1aflox allele and Mob1adelta allele, respectively, are shown.
