Caption Disruption of the murine Fbx15 gene. A, embryonic stem cells containing a gene-trapped (GT) Fbx15 allele were used to generate the Fbxl5Gt(OST386421)Lex allele. A trapping vector containing a promoter-less neomycin (Neo) resistance cassette flanked by a splicing acceptor (SA) site and a polyadenylation signal sequence (pA) was inserted 3' of Fbx15 exon 2. Utilization of the SA site generates a truncated FBXL5 mRNA transcript competent for Neo expression. The trapping vector also contains the first exon of the Bruton's tyrosine kinase (Btk) gene flanked by the PGK promoter sequence and a splicing donor site (SD). The resultant chimeric fusion transcript was used to generate a sequence tag of the trapped gene by 3' RACE.
