Caption Construction of the Snai3tm2.1Grid and Snai3tm2.2Grid alleles. Schematic representation of the wild-type Snai3 allele, the targeting vector, and the targeted allele in which an FRT-neo-FRT-loxP- cassette was inserted between exons 1 and 2, and the second loxP site was inserted 5' to exon 1. Removal of exon 1 and the neo cassette via cre-mediated recombination results in the Snai3tm2.2Grid allele. Removal of the neo cassette via flp-mediated recombination leaves exon 1 floxed and generates the Snai3tm2.2Grid allele. Exons are indicated by rectangles with coding sequences designated by gray shading, and noncoding sequences in black. A diphtheria toxin (DT) cassette was included for negative selection of randomly integrated clones. Hybridization probes used for Southern blot analyses are indicated.
