Caption A. Generation of the Ptprgtm1.1Lex allele. Targeting strategy used to introduce the point mutation into the Ptprg locus. Homologous recombination (represented by X) between the targeting vector and the Ptprg gene results in the replacement of a G residue with a C resulting in a Cysteine to Serine amino acid change. The LoxP flanked Neo cassette is excised during the chimeras breeding step.
