Generation of the Hltf^tm1.1Geno and Hltf^tm1.2Geno alleles. Panel A, recombined allele in F1 mice, i.e. FRT-flanked (double red triangles) Neo resistance selection cassette is downstream of exon 12, and two loxP sites (blue triangles) are upstream of exon 11 and downstream of exon 12. Exons are numbered green rectangles. Next, the floxed allele, Hltf^tm1.1Geno, in F2 mice results from breeding F1 mice with Flp recombainse expressing Flp deleter mice to remove the Neo resistance cassette. Lastly, the knockout allele, Hltf^tm1.2Geno, (F3 mice) results from breeding F2 heterozygous Neo-excised mice to mice expressing Cre recombinase under the control of the CMV promoter.
