Caption Generation of the Cep290Gt(CC0582)Wtsi allele. (A) The murine Cep290 gene measures 85 kb and extends over 53 exons. The Cep290 start codon is located in exon 1, and the stop codon is located at exon 53. The encoded protein contains numerous predicted coiled-coil (CC) domains as well other predicted domains, including Tropomyosin (TM, blue), KID (red), bipartite nuclear localization signal (NLS-BP, green), P-loop (purple), and Structural Maintenance of Chromosomes Homology (SMC Homology, orange). Predicted domains were obtained from Ensembl (www.ensembl.org). (B) Zoom in of gene trap insertion site with oligonucleotide primer annealing sites F4, R2, and R5 shown. F4 targets the start of exon 23 in the Cep290 gene. The gene trap is located in the intron between exon 23 and exon 24, leading to a predicted truncated protein beyond exon 23. For genotyping, a reverse oligonucleotide primer R5 was used to amplify a 300-base pair (bp) region containing a gene-trap sequence whereas a reverse oligonucleotide primer R2 (product size 550 bp) was used to identify wild-type mice.
