Caption | Knock-in of E160D Fen1 mutant in mouse germ line to generate the Fen1tm3.1Bhsh allele. (a) The expected restriction maps of E160D targeting vector and different types of Fen1 alleles. A DNA fragment containing the mouse G480T Fen1 mutation (E160D in amino acid sequence) was generated by site-directed mutagenesis using primers mE160DF and mE160DR. It was then ligated into the gene targeting vector, PKO scrambler NTK (Invitrogen). Two Lox P sequences flank the neomycin selection marker, which was removed by transient expression of Cre-recombinase in the mutant ES cells (Fukushige, 1992, Proc. Natl. Acad. Sci. U S A 89, 7905-7909). The remaining copy of the Lox P sequence is located in the intron sequence 200 bp upstream of the ATG starting codon of Fen1 open reading frame. The c11orf10 gene (RIKEN 1810006k21), which is located immediately upstream of Fen1 in a reverse orientation (Adachi, 2002 Omics, 6, 273-279) was not affected by the insertion of the Lox P sequence. The insertion site is 2.33 kb away from the c11orf10 gene. | ||||
