The EUCOMM Dmxl2 gene trap allele with conditional potential and the Dmxl2 allele after the action of Cre recombinase. (A) The Dmxl2 gene trap allele (tm1a allele) contains an IRES:LacZ trapping cassette with an acceptor splice site (En2 SA) and a floxed neoR cassette. LoxP sites flank the critical exon 7 and the neoR cassette. FRT sites flank the IRES:LacZ and neoR cassettes (adapted from www.knockoutmouse.org/about/eucomm). The IRES:LacZ trapping cassette and the floxed neoR cassette have been deleted by the FLIP recombinase to generate Dmxl2lox/lox (Dmxl2tm1c(EUCOMM)Wtsi allele) mice. The critical exon 7 was then deleted by the Nestin cre-recombinase to generate both Dmxl2lox/wt and nes-Cre;Dmxl2-/wt. Dashed lines indicate possible RNA splice isoforms.
