| Caption | Generation of the Cd209btm1.1Cfg allele. (a) Schematic representation of the targeting strategy. A targeting vector was constructed to flank a 4.4-kb PCR amplified BstBI fragment (red) including exons 1-4 of the Cd209b (SIGNR1) gene with two loxP sites. Another loxP site was inserted to flank TK and PGKNeo. A PCR-amplified 3.0-kb Not I- Sac II fragment (green) and a 3.4-kb XhoI-KpnI fragment (blue) were used as 5' and 3' homologous regions, respectively. ES clones with floxed-TK-Neo alleles were selected. Cre-mediated excision was performed to generate floxed and delta (Cd209btm1.1Cfg) alleles. | ||||
