| Caption | Generation of the Lyve1tm1.1(EGFP/cre)Cys allele. Map of the construct and targeted locus. (A) A 5.4-kb and a 3.4-kb fragment homologous to the 5' and the 3' end, respectively, of the 3' untranslated region (UTR) was PCR amplified and subcloned into the pIRES-GFP-hCre-FNF vector using an In-Fusion Dry-Down PCR cloning kit (BD). The resulting construct was linearized by SacII and electroporated into E14 embryonic stem cells. Clones were screened by 3' end long PCR using a Long Template PCR system (Roche). | ||||
