Caption Conditional targeting of the ROSA26 locus to generate various reporter lines. The black box indicates non-coding exon1 of ROSA26 genomic locus and black triangles indicate loxP sequences. MC1-DT-A is a diphtheria toxin A fragment gene for negative selection. Probe represents the probe that used for Southern blot analysis. AflII is the restriction enzyme site for Southern blot analysis. P1 and P2 are primers for PCR screening of homologous recombinant ES cells. P3, P4, P5, and P6 are PCR primers for routine genotyping of mutant mice and embryos. SA, adenovirus splice acceptor sequence; tpA, triple repeats of SV40 polyadenylation sequence; bpA, bovine growth hormone polyadenylation sequence.
