The 5' and 3' regions and introns are indicated by the striped bars for the mutant G85R. The coding sequence for EYFP (Clontech) was fused in frame at the last codon of SOD1 in exon V with a short linker. The G85R mutation was introduced into exon IV by oligonucleotide-directed mutagenesis of a restriction fragment, which was then substituted for the wild-type fragment.
