Caption Targeting of the mouse Lyl1 genomic locus. (a) Schematic representation of the Lyl1 genomic locus. Map and restriction enzyme sites demonstrate the strategy used for targeting and screening ES clones. Thick lines indicate the DNA sequence used for homologous recombination, the black arrow indicates the PGK promoter that drives the neomycin resistance gene, and the grey arrows indicate the loxP sites. The diphtheria toxin gene (DTA) was used for negative selection. BamHI and BglI sites were used for screening the 3' and 5' regions, respectively.
