(A) Structure of the targeting vector and chromosomal organization of the Cdc25A locus before and after Cre-mediated excision. The genomic organization of the mouse Cdc25A gene was disrupted by insertion of neomycin phosphotransferase cDNA driven by the phosphoglycerine kinase promoter (pGK-neo) as a selectable marker into intron 3. Exons 1-5 are represented by black boxes. The location of BstXI (B) and XbaI (X) sites are indicated; loxP sites are represented by yellow triangles. The sizes of the upstream (7.5 kb) and downstream (1.2 kb) homologous arms are indicated. The red bars indicate positions of the probes used for Southern blot analysis (internal and 3' probe).
