| Caption | Generation of the Khdc3tm1Dean allele. (A) Schematic representation of the normal (chr 9:72950,25672,952,250) and null allele after targeting with a construct containing positive (PGK-Neo) and negative (MC1Tk) selectable markers and 3.1-kb and 5.6-kb homologous arms (thicker lines) that are 5' and 3', respectively, to the Neo cassette. The 5' (511 bp, 353 bp outside of the targeting construct) and 3' (428 bp, 732 bp outside of the targeting construct) probes were used for Southern hybridization. PCR genotyping was done using primers P1 and P2 for the normal allele and primers P1 and P3 for the null allele. Arabic numbers indicate exons (light-blue boxes). Arrows indicate cleavage sites of Bcl1 restriction enzyme. | ||||
