Caption Panels A-C: Fgfr2tm1Schl/Fgfr2+ (Fgfr2cfC324Y/+) (B) mice are characterized by protruding eyes, rounded cranium and severe reduction in the development of the midfacial area. +/+ wildtype (A), Fgfr2tm2Schl/Fgfr2+ (Fgfr2cCLR/+) (C). Panels D-F: The skulls of +/+ wildtype (D) Fgfr2tm1Schl/Fgfr2+ (E) and Fgfr2tm2Schl/Fgfr2+ (F) were stained with alizarin red. All sutures are visible (arrows) in the wildtype and Fgfr2tm2Schl/Fgfr2+, however in the Fgfr2tm1Schl/Fgfr2+ the cornal sutures are fused with significantly shortened facial region. Panels G-I: The wildtype (G) and Fgfr2tm2Schl/Fgfr2+ (I) have fully opened coronal, sagittal and lambdoid sutures. In the Fgfr2tm1Schl/Fgfr2+ (H) the coronal sutures are completely fused on both sides of the skull (arrows). Panels J-L: Coronal cross-sections of 6 week old mice. The coronal sutures of the wild type (J) and Fgfr2tm2Schl/Fgfr2+ (L) remain open whereas the Fgfr2tm1Schl/Fgfr2+ are fused (K) (arrows). Panels M-O: Histological analysis of 1 week old mice show the presence of bone trabeculae in the sutural mesenchyme in the Fgfr2tm1Schl/Fgfr2+ (N) and the same sutures are unaffected in the wildtype (M) and Fgfr2tm2Schl/Fgfr2+ (O). Abbreviations: Coronal suture (C); sagittal suture (S); Lambdoid duture (L); frontal suture (F); frontal bone (fb); parietal bone (pb); ossification front (of); mesenchyme (me); bone trabecula (Tb). Scale bars: 2mm (D-I) 100um (M-O)
