Caption | A: Representative hematoxylin and eosin stained cross-sections of an E16.5 Myh10tm6(Myh9/GFP)Rsad/Myh10tm6(Myh9/GFP)Rsad (Ba*/Ba*) mouse heart showing ventricular septal defect (VSD, yellow arrow in right panel) which is not seen in wild type (B+/B+) mice. RA, right atrium; LA, left atrium; RV, right ventricle; LV, left ventricle; VS, ventricular septum. Scale bar, 0.4 mm. B: Representative image of binucleated and enlarged cardiac myocytes in an E13.5 mutant mouse heart. Yellow arrows in the mutant heart indicate binucleated cardiac myocytes. Scale bar, 50 um. C: Representative flow cytometry profiles of cells isolated from E16.5 hearts. The cells were stained with an anti-alpha-actinin antibody to identify the cardiac myocytes (y axis). The percentages of cardiac myocytes and non-cardiac myocytes in wild type mice are 64.6 +/- 1.3 and 35.4 +/- 1.3% (mean +/- S.E., n = 4), respectively; whereas in mutant mice, the percentage is 38.0 +/- 6.9% for cardiac myocytes and 62.0 +/- 6.9% for non-cardiac myocytes (mean +/- S.E., n =6). Note that the GFP signal is detected in both myocytes and non-myocytes in the hearts of mutant mice (x axis). | ||||
