Caption | Histological and histochemical analysis of skeletal muscles. Fresh frozen gastrocnemius muscle specimens from wild-type (A,D,G and J), Ldb3tm2Chen/Ldb3tm2Chen (Cypher2sKI, B,E,H and K) and Ldb3tm3Chen/Ldb3tm3Chen (Cypher3sKI, C,F,I and L) mice were stained with hematoxylin and eosin (A-C), modified Gomori trichrome stain (D-F), or reacted for myofibrillar ATPase, pH 4.3 (G-I) and cytochrome c oxidase (J-L) activity. Variability in myofiber size and numerous atrophic fibers were observed in Ldb3tm2Chen/Ldb3tm2Chen (B) and Ldb3tm3Chen/Ldb3tm3Chen (C) muscle when compared with wild-type littermates (A). The black arrows in Ldb3tm2Chen/Ldb3tm2Chen (B and E) and Ldb3tm3Chen/Ldb3tm3Chen (C and F) muscle indicate subsarcolemmal invaginations resulting in a lobulated pattern, which are basophilic with the hematoxylin and eosin stain and reddish with Gomori trichrome stain. The white arrow in Ldb3tm3Chen/Ldb3tm3Chen (F) indicates regions within the muscle tissue with a clear, hyaline appearance, which were associated with a loss of cytochrome C oxidase activity (L). Note that lobulations and translucent regions were not apparent in wild-type muscle (A,D,G and J). The atrophic fibers, lobulations, and clear areas were present in both muscle fiber types (I).
