Caption A, degeneration of neurons is evident only in the brain of a Npas4tm1Ooe/Npas4tm1Ooe (KO) mouse (30 weeks old) with the Fluoro-jade B fluorescence reagent, known to specifically stain degenerated neurons. B, anti-glial fibrillary acidic protein (GFAP) immunostaining. Only three pickup examples of hippocampus (KO11-13, 30 weeks old) and one pickup cerebral cortex (KO-11, 30 weeks old) are shown with the control wild-type (Wild) littermate. Note GFAP induction in the hippocampus or the cerebral cortex of mutant mice accompanied by glial cell activation, suggestive of proximal neuronal cell damage, with large individual differences in the sites and extent. C, Immunostaining results for a mutant mouse (KO-11) with several brain damage marker antibodies. Top panel: positive TUNEL of apoptosis in neurons of the hippocampus (CA1-2 region). Middle panel: staining with the anti-IbaI antibody, which specifically reacts with microglial cells, shows assembly of microglia in the hippocampus. Bottom panel: anti-nitrotyrosine antibodies react with oxidative species only in degenerated neurons in the mutant brain (cerebral cortex).
