(a) A control E10.5 embryo shows normal forelimb (arrow) and heart (clear arrowhead) morphology, whereas a Rspo3tm1.2Jcob/Rspo3tm1.2Jcob (Rspo3-/-) embryo (b) has obvious vascular defects, including hemorrhaging (black arrowhead) and a swollen pericardial sac (clear arrowhead). The small forelimb bud (arrow) is characteristic of an early E10 embryo. (c, d) Control and mutant E10.75 embryos within yolk sac membranes. The control membrane shows obvious vascularization (arrows) while the mutant is pale (arrow) without visible blood vessels. (e, f) The E10.75 embryos from (c) to (d) after removal of embryonic membranes. The mutant (f) has an engorged pericardial sac (clear arrowhead) and small forelimb buds (arrow) and has the overall size of an early E10 embryo. A weak heartbeat was detected in this embryo. (g) A control embryo shows normal E11.5 morphology. A mutant littermate (h) is dead and shows developmental arrest at early E10, as judged by embryo size and forelimb bud morphology (arrow) (compare to (b, f)).
