Tli1 resistance/susceptibility

Significant differences in the susceptibility to MNU-induced T-lymphoma was observed between the inbred mouse strains AKR/J and BALB/cJ. 180 days after MNU injection, 83% of AKR/J mice and 37.5% of BALB/cJ mice developed T-lymphoma. Tumor incidence was adominant trait as 93% of F1 mice developed T-lymphoma. Further, the final tumor incidence of Tli1 mice homozygous for the BALB/cJ allele is significantly lower (P = 0.0004) compared to the tumor incidence of the total F2 population. The authors propose that Tli1 is a recessive resistance gene for MNU-induced T-lymphoma in BALB/cJ mice. Cumulative results of a genome-wide screen from 285 F2 mice identified a locus Til1 in a 10 Cm interval on central mouse Chromosome 1 between D1Mit87 and D1Mit423. Inheritance of parental alleles at the markers D1Mit87, D1Mit217, D1Mit193, D1Mit30 and D1Mit423 deviate from the expected values with associated P values close to or less than 0.001. the likely location of Tli1 is the segment spanning the region from ~65 cM to~75 cM on central mouse Chromosome 1. This location was confirmed by results obtained from the analysis of a BALB/c.AKR/J-Tli1 congenic strain carrying AKR/J alleles on a region of mouse central Chromosome 1. 73.8% of the congenic strain developed T-lymphomas upon MNU injections. This differed significantly from the tumor incidence in the parental BALB/cJ mice (P = 0.0007) and confirmed the presence of Tli1 on central mouse Chromosome 1. Two candidates from the Tli1 are the Il10 and Cmkar4 genes.
