The genetic basis for differential sensitivity of inbred mice to inflammatory bowel disease induced by dextran sulfate sodium (DSS) was investigated by crossing susceptible C3H/HeJ mice to partially resistant C57BL/6J mice. 86 (C3H/HeJ x C57BL/6J)F2 mice and 90 [(C3H/HeJ x C57BL/6J) x C57BL/6J]N2 backcross mice were analyzed for the presence of QTLs influencing DSS-induced colitis. An additive QTL on mouse Chromosome 5 between D5Mit188 and D5Mit285 (susceptibility marked by the B6 allele at D5Mit216) was significantly linked to several of the colon subphenotypes like severity, hyperplasia, ulceration, and % area involved. The confidence level for linkage of QTL to markers on Chromosome 5 exceeded the threshold for declaring significant linkage (P = 0.0000137). Possible candidate genes in the region include Selp1 and Pdgfa.
