Congenic strain C57BL/6J.SPRET-Hprta carries 17 cM of M. spretus-derived mouse Chromosome X (from DXMit48 to DXMit60) on a C57BL/6J background and exhibits low fertility weight and low testes weight. A locus responsible for this phenotype, Ihtw1 (interspecific hybrid testis weight 1), has been mapped to a resolution of 1.8 cM on mouse Chromosome X at approximately 17.05 cM - 19.0 cM. Mapping was conducted on 15 subcongenic lines generated from (C57BL/6J.SPRET-Hprta x C57BL/6J)F1 x C57BL/6J recombinant backcross animals. Markers DXMit23 and DXMit73 flank the smallest common interval associated with low fertility and low testes weight in the subcongenic lines. Ihtw1 maps near Hprt, Zic3, Fgf13, and Tnfsf5. An EST, DXWsu72e, also mapped to the locus. Gpc4 maps approximately 1 cM proximal to Ihtw1. It is possible a second locus exists on mouse Chromosome X in the 17 cM congenic region that is required for the full phenotype. The authors tentatively name this locus Ihtw2.
