100 (C57BL/6 x NOD)F2 animals were treated with gamma-irradiation and assessed for T-cell apoptosis. Linkage analysis was conducted using markers near previously identified Idd (insulin dependent diabetes susceptibility) loci to find association between diabetes and resistance to T-cell apoptosis. T-cells harvested from NOD animals are resistant to experimentally induced apoptosis whereas T-cells from C57BL/6 animals are susceptible to experimentally induced apoptosis. Significant linkage to apoptosis resistance to was detected in a 3 cM region on mouse Chromosome 1 from 30.5 cM - 32.8 cM overlapping Idd5 (P = 0.000086 at D1Mit21). Authors name this locus Capop (cyclophosphamide induced apoptosis) and confirmed the locus in congenic strain NOD.B6-C1.1 carrying the NOD-derived Idd5 on a C65BL/6 background. NOD.B6-C1.1 exhibits resistance to induced T-cell apoptosis similar to parental strain NOD. Genes mapping near Capop include Ctla4, Cd28, Cflar, Casp8, and Icos. Cd152 deficient mice show similar resistance to gamma-irradiation induced T-cell apoptosis as NOD mice and is a likely candidate gene for Capop and/or Idd5.
