Animals from a (B10.S/DvTe x SJL/J)F1 x B10.S/DvTe backcross were used to map loci involved in experimental allergic encephalomyelitis (EAE) susceptibility. Animals were assessed for EAE susceptibility and severity either with or without pertussis toxin (PTX+ or PTX-) challenge. Previously identified loci Eae9, Eae7, and Eae11 were detected in this study and 3 new loci, Eae14, Eae17, and Eae25, were identified. Eae14 was identified in PTX- animals and maps to mouse Chromosome 8 spanning 10 cM - 33 cM with a maximum LRT score of 25.9 at D8Mit190. B10.S-derived alleles appear to affect EAE susceptibility, severity, and spinal chord (SC) demyelination at this locus. Potential candidate gene mapping near Eae14 are Gsr and Casp3. Eae17 was identified in PTX- female animals and maps to mouse Chromosome 10 spanning 10 cM - 33 cM with a peak LRT score of 16.2 at D10Mit31. SJL/J-derived alleles are associated with EAE severity at this locus. Potential candidate genes for Eae17 are Mmp11, Timp3, Igf1, Mif, and Aire. Eae25 was identified in PTX- male animals and maps to mouse Chromosome 18 with a maximum LRT score of 20.4 at approximately 54 cM. SJL/J-derived alleles are associated with spinal chord demyelenation and monocyte/lymphocyte infiltration at Eae25. Candidate genes mapping near Eae25 include Mc2r and Mbp.

Editor's note: Author's symbol Eae18 was reassigned to Eae25 in MGI.
