Linkage analysis was performed on CXB (C = BALB/cBy, B = C57BL/6J) recombinant inbred (RI) strains to identify loci associated with susceptibility to Neuroadapted Sindbis virus (NSV) infection. Progenitor strain BALB/cBy is resistant to infection by NSV whereas progenitor strain exhibits C57BL/6J 100% mortality from encephalomyelitis post-infection. A suggestive QTL linked to NSV viral RNA levels was detected on mouse Chromosome 2 from 65.2 cM - 71 cM and mapped in a (C57BL/6J x BALB/cBy)F2 intercross as highly significant (LRS = 16 at D2Mit447). C57BL/6J-derived alleles contribute to increased viral RNA levels in female animals at this locus, designated Nsv1. Nsv1 maps near Fkbp7, Fshb, Kcna4, Snap23, Blvra, Chrm5, Cat, Rag1, Rag2, and Bdnf. Bdnf is being investigated as a possible candidate gene for Nsv1. A suggestive locus mapped to mouse Chromosome 7 near D7Mit223 but was not able to be confirmed in the F2 intercross.
