• Experiment
  The authors used previous genetic and physical mapping studies to refine the Lp interval to a critical region of ~ 450kb. The complete transcript map was developed using a combination of computational methods. The order of markers was determined to be: D11Mit113 - Cd84 - Vangl2 - 340L6T7 - Nhlh1 - D1Sta2 - Ncstn - Copa - Pex19 - Timm23 - D1Ucla4 - Pea15 - Casq1 - 446G11T7 - D1Sta3 - Atp1a4 - 423N13T7 - 378C5T7 - Atp1a2. Comparative sequence analysis identified a G -> A transition in exon 8 of the Ltap gene. This substitution was unique to the Lp mutant allele and was not seen in 8 other mouse strains. The substitution caused a change from serine to asparagine and this is beleived to disrupt the normal function of the Vangl2 protein.
