Genome scan at an average marker resolution of 23 cM was performed on an outcross population of 253 (BALB/cJ x C57BL/6J)F1 x (C3H/HeJ x DBA/2J)F1 animals to identify loci associated with long life span. Animals from the first 20% of deaths were excluded from the data set to maximize identification of longevity loci. Lifespan1 mapped to 48 cM on mouse Chromosome 9 with p<0.01 at D9Mit110. C3H/HeJ-derived alleles confer long life in male animals at this locus. Lifespan2 mapped to 35 cM on mouse Chromosome 10 with p<0.05 at D10Mit15. DBA/2J-derived alleles confer long life in males at Lifespan2. Lifespan3 mapped to 52 cM on mouse Chromosome 12 in linkage to long life in both males and females with p<0.01 at D12Mit167. The C57BL/6J + C3H/HeJ heterozygous genotype confers increased life span at this locus. Lifespan1 and Lifespan2 appear epistatic-80% of male animals exhibiting the greatest longevity inherited both the C3H/HeJ allele at Lifespan1 and the DBA/2J allele at Lifespan2. Two suggestive loci with additive interactions mapped to D16Mit182 and D2Mit58. BALB/cJ-derived alleles at D16Mit182 and C3H/HeJ-derived alleles at D2Mit58 additively confer longer life in female animals.
