A genome scan using 75 polymorphic markers was performed on a population of (NZW/LacJ X PL/J)F1 x NZW/BlNJ animals to identify loci contributing to systemic lupus erythrematosus (SLE). Inbred strains NZW/LacJ and NZW/BlNJ are susceptible to SLE whereas PL/J is resistant. Significant linkage to mortality and proteinuria was detected on distal mouse Chromosome 2 at approximately 86 cM with a LOD score of 6.6 at D2Mit285. This locus was named Rends (author's designation: Wbw1). Rends has a QTL range of 58 cM - 93 cM and is associated with the NZW/LacJ-derived susceptibility allele. PL/J-derived alleles at Rends appear to exhibit a dominant protective effect. A possible candidate gene for Rends is Il1. Defects in Il1 expression have been observed in 2 mouse models of SLE. Rends appears to interact with NZW/LacJ-derived alleles to increase disease severity at suggestive loci on mouse Chromosome 17 (24 cM) near D17Mit177 (which the authors designate Wbw2) and distal mouse Chromosome 1 near D1Mit17 (106 cM). The Chromosome 1 locus maps near a previously identified SLE susceptibility QTL, Sle1c. A possible candidate gene identified for this region is Cr2.
