    Linkage analysis was performed on 28 C57BL/6J x DBA/2J (BXD) recominant inbred (RI) strains to identify loci associated with hematopoietic response to early cytokines. Primitive progenitor cells of parental strain C57BL/6J exhibit stronger proliferation in response to flt3L, KL, and TPO compared to parental strain DBA/2J. Also, the absolute number of Lin-Sca1++ cells is 2-fold higher in C57BL/6J than in DBA/2J. A major locus, Hrec, mapped from 73.2 cM (D2Mit495) - 77.6 (D2Mit411) cM on mouse Chromosome 2with LRS = 18.8 for proliferative response and LRS = 9.4 for absolute number of Lin-Sca1++ cells. C57BL/6J-derived alleles confer increased proliferation and number of Lin-Sca1++ cells at Hrec whereas DBA/2J-derived alleles confer decreased proliferation and cell numbers. This locus was confirmed in congenic strain B6.D2-(D2Mit133-D2Mit200) which carries a DBA/2J-derived segment of chromosome 2 DNA from 66.8 cM - 107 cM on a C57BL/6J genetic background. This congenic exhibits significantly fewer Lin-Sca1++ cells compared to the C57BL/6J background strain thus confirming the decreasing effect of DBA/2J-derived alleles at Hrec. Possible candidate genes for Hrec are Jag1, Bmp2, Il1a, and Il1b. A highly suggestive QTL linked to Lin-Sca1++ cell number mapped to approximately 79 cM on mouse Chromosome 4 (D4Mit33 and D4Mit343) with LRS = 15.9. DBA/2J-derived alleles confer increased cell numbers at this locus.
