    BALB/c mice develop Th2-biased immune responses with enhanced airway hyperreactivity (AHR) whereas DBA/2 mice develop reduced Il4 responses that protect them from the development of AHR. Congenics and backcross analysis were used to identify a genetic locus on chromosome 11, Tapr, that appeared to regulate both Th2 cytokine production and AHR. More than 2000 N2 mice from the backcross were genotyped and phenotyped to map the relevant locus within the proximal congenic region between D11Mit135 and D11Mit260. 22 additional markers were then identified and used to provide a higher resolution map of the region. Further genetic and cytokine analyses were used to show that the genetic locus controlling high Il4 responses is located between D11Mit271 and D11Mit22. In addition, high Il4 production was observed in mice with the Balb/c allele at Havcr1. Thus Tapr was non recombinant with Havcr1. Based on the frequency of recombinant haplotypes between D11Mit271 and D11Mit22, the Tapr locus was mapped to a 0.3-0.5 cM region. This region is homologous to regions on human Chr 5q that are linked to human atopy and asthma. Subsequent analyes with AHR phenotypes also showed that the locus associated with AHR is also located within D11Mit271 and D11Mit22 and is non-recombinant with Havcr1. This suggests that the same locus Tapr is responsible for both Il4 regulation and AHR.
