Notes
  • Experiment
58 animals from either a (C57BL/6J x Swiss Webster)F2 intercross or a (C57BL/6J x Swiss Webster)F1 x Swiss Webster backcross were screened for 61 polymorphic markers (average resolution = 30 cM) to detect QTLs associated with MPTP-induced neuron loss. Parental strain C57BL/6J is susceptible to MPTP-induced neuron loss of the substantia nigra while parental strain Swiss Webster is resistant. A QTL named Mptp1 on mouse Chromosome 1 was identified with LOD=4.5 and a QTL range of 93.3 cM - 109.6 cM between D1Mit113 and D1Mit293. Surprisingly, C57BL/6J-derived alleles confer resistance to MPTP-induced neuron loss with a dominant mode of inheritance. Homozygosity for Swiss Webster-derived alleles at Mptp1 is associated with increased neuron loss.
