218 animals from a (CBA/J-Pde6brd1/Pde6brd1 x C57BL/6J-Pde6brd1/Pde6brd1)F2 intercross were typed for 74 microsatellite markers at an average resolution of 15 cM to identify QTLs associated with circadian photosensitivity. Parental strain C57BL/6J-Pde6brd1/Pde6brd1 exhibits a greater phase shift of circadian rhythm locomotor activity when subjected to a monochromatic light pulse compared to parental strain CBA/J-Pde6brd1/Pde6brd1. Two significant QTLs were identified on mouse Chromosomes 12 and 17. Circp1 maps to 6 cM on Chromosome 12 with a LOD score of 3.5 at D12Mit170. CBA/J-derived alleles confer increased photosensitivity with recessive inheritance at Circp1. Circp2 maps to 10.5 cM on Chromosome 17 with a LOD score of 3.7 at D17Mit113. CBA/J-derived alleles confer increased photosensitivity with dominant inheritance at Circp2. A possible candidate gene for Circp2 is Junb. Further analysis revealed that Junb expression is significantly reduced in CBA/J-Pde6brd1/Pde6brd1 animals after exposure to light. Suggestive loci were also identified at 39.3 cM (D8Mit75, LOD=2.2) and 44 cM (D8Mit242, LOD=2.2) on mouse Chromosome 8, and at 23 cM (D12Mit54, LOD=2.7) on mouse Chromosome 12.
