    Genome scan at a resolution of 10 cM was performed on 215 (NZB x NZW)F1 x NZW backcross animals to identify QTLs associated with anxiety-related behavior. Parental strain NZB exhibits increased anxiety as measured by the decreased number of open arm entries in the elevated plus maze test compared to parental strain NZW. (NZB x NZW)F1 hybrid animals show an even greater decrease in the number of open arm entries compared to anxiety prone parental NZB.

    Anxty, a QTL showing significant linkage to anxiety, mapped to 49.6 cM on mouse Chromosome 4 with LOD=3.2 at D4Mit119. The QTL range of Anxty spans from 37.5 cM (D4Mit142) to 59.1 cM (D4Mit16). NZB-derived alleles confer increased anxiety (decreased number of open arm entries) in an additively-inherited manner. This locus was confirmed by constructing a congenic line heterozygous for NZB/NZW-derived alleles at Anxty (20 cM region) on a NZW genetic background. As expected, the congenic animals displayed increased anxiety in the elevated plus maze test. A candidate gene mapping to the Anxty region is Ifna. This candidate gene is particularly attractive because humans and mice administered Ifna exhibit increased anxiety. Expression analysis revealed a slight but significant increase in Ifna expression in the brains of NZB and (NZB x NZW)F1 animals compared to NZW.

    Suggestive linkage to anxiety was detected at 65.5 cM on mouse Chromosome 2 with LOD=1.3 at D2Mit254. The NZW-derived allele confers increased anxiety (decreased number of open arm entries) in an additively-inherited manner.

    Animals heterozygous at Anxty and homozygous for NZW-derived alleles at D2Mit254 display the lowest number of open arm entries.

    Suggestive linkage to locomotor activity (as measured by the number of closed arm entries in the elevated plus maze test) was detected at centromeric mouse Chromosome 19 (LOD=2.0).
