The epilepsy QTL El2 maps to 57 cM on mouse Chromosome 2. A congenic strain named ABP/EL-El2e/Frk was constructed by introgressing an EL-derived portion of chromosome 2 containing the El2 locus (approximately 42 cM - 96 cM) onto the genetic background of ABP/LeJ. Parental strain EL is susceptible to seizures whereas parental strain ABP/LeJ is resistant. A series of nested subcongenic lines was subsequently derived from ABP/EL-El2e/Frk by backcrossing to the ABP/LeJ parent and selecting for recombinants. A total of 8 subcongenic lines were constructed, 4 carrying overlapping region of proximal El2 and 4 carrying overlapping regions of distal El2.

None of the subcongenic lines fully recapitulated the rhythmic tossing seizure phenotype of ABP/EL-El2e/Frk. Data suggests there are at least 2 genes within the El2 region affecting sensitivity to seizures with possible sex influence effects. One gene resides in the proximal (centromeric) half of the El2 interval between D2Mit7 (28 cM) and D2Mit41 (49 cM) and the other gene resides in the distal (telomeric) half between D2Mit58 (51.4 cM) and D2Mit30 (69 cM). It appears that the gene in the centromeric half of the El2 locus influences susceptibility to PZT-induced seizures.
