    The Fob3 locus was identified at 34 cM on mouse Chromosome 15 by Horvat et al, 2000 (J:59227) in F-Line (fat) and L-Line (lean) mice. In the current study the Fob3 interval was further dissected using recombinant progeny testing and congenic lines. Congenic animals were created by introgressing the Fob3 interval from L-Line animals onto the F-Line genetic background. Three Fob3 congenic lines were created with the following L-Line-derived Chr15 intervals: A congenic (D15Mit150 [12 cM] - D15Mit107 [49 cM]), B congenic (D15Mit184 [25 cM] - D15Mit39 [56 cM]), and D congenic (D15Mit184 [25 cM] - D15Mit107 [49 cM]). Sixty microsatellite markers were screened in 989 recombinant progeny and grandprogeny.

    The effect of Fob3 (via fat percentage) was confirmed in the congenic animals. Two separate regions within the Fob3 interval showed linkage to growth traits at approximately 27 cM and 68 cM. These loci were named Fob3a and Fob3b, respectively, and appear to participate in epistatic interactions. The interval of Fob3a spans 22 cM -32 cM and the interval of Fob3b spans 44 cM - 70 cM. Fob3a is a late onset obesity locus exerting an effect after 42 days of age and shows strongest linkage to relative gonadal fat pad weight, fat percentage, and body weight at 42 and 70 days of age. Fob3b is an early onset obesity locus with an effect observable at 21 days of age and shows strongest linkage to fat percentage, and body weight at 21, 42, 70, and 98 days of age.

    Previously identified growth trait QTLs mapping near Fob3a are Bdln2 (15 cM), Dob3 (22.8 cM), and Dob4 (18.35 cM). Candidate genes for Fob3a include Cox6c and Trhr (24.7 cM). Candidate genes for Fob3b include Ndufb9, Sqle, Rhpn1, Oc90 (37.5 cM), Myc (32 cM), Dgat1 (46.9 cM), and Tef (46.7 cM).
