    Linkage analysis was performed on male animals from a series of Recombinant QTL Introgression (RQI) strains (derived from BALB/cJ or CXBI donor DNA on a C57BL/6ByJ genetic background) to identify QTLs associated with alcohol consumption. Progenitor strains BALB/cJ and CXBI exhibit lower ethyl alcohol consumption (EAC) compared to progenitor strain C57BL/6ByJ. All of the RQI strains exhibit greater EAC compared to BALB/cJ and CXBI. 97 microsatellite markers at an average spacing of 4.91 cM were used for linkage analysis. Several provisional QTLs were identified.

    Linkage to EAC mapped to 6.5 cM on mouse Chromosome 1 near D1Mit167 (P<0.01). There was a significant phenotypic difference between RQI strains carrying homozygous BALB/cJ-derived alleles at D1Mit167 and background strain C57BL/6ByJ. Oprk1 maps near this locus and is considered a potential candidate gene. This locus colocalizes with a previously mapped alcohol preference QTL named Alcp4 (8.3 cM).

    A suggestive locus mapped to 102 cM on mouse Chromosome 1 near D1Mit221 (P<0.05). Homozygosity for BALB/cJ-derived alleles at D1Mit221 confer increased alcohol consumption compared to homozygosity for C57BL/6ByJ-derived alleles. A potential candidate gene for this locus is Tgfb2 at 101.5 cM.

    Linkage to EAC was also detected at 107 cM on mouse Chromosome 2 near D2Mit74 (P<0.01). Oprl maps near this locus (110 cM) and is considered a potential candidate gene.

    Authors report these EAC loci as provisional and require further study and confirmation. Together these 3 loci account for approximately 32% of the genetic variance.
