Linkage analysis was performed on 13 CXB (C=BALB/cBy; B=C57BL/6J) recombinant inbred (RI) strains to identify QTLs associated with susceptibility to lung tumor development and lung inflammatory response induced by various chemicals. Parental strain BALB/cBy exhibits higher incidence of urethane- and BHT-induced lung tumors compared to C57BL/6J. BALB/cBy also exhibits increased inflammatory response compared to C57BL/6J. Authors report the following loci as provisional and requiring further study and confirmation.

    Loci at 22 cM (D3Mit306) - 25 cM (D3Mit5) and 76.2 cM (D3Mit86) - 87.6 cM (D3Mit91) on mouse Chromosome 3 showed linkage to lung inflammatory response (LOD=3.3 and 3.6, respectively).

A locus on mouse Chromosome 4 from 7.5 cM (D4Mit95) - 12.1cM(D4Mit236) showed linkage to lung inflammation (LOD=2.4), while a locus at 60 cM (D4Mit178) showed linkage to U/BHT-induced lung tumors (LOD=2.1).

A locus spanning 43 cM (D8Mit240) - 45 cM (D8Mit312) on mouse Chromosome 8 showed linkage to lung inflammatory response (LOD=6.8).

A locus at 61 cM (D9Mit16/D9Mit20) on mouse Chromosome 9 showed linkage to lung inflammatory response (LOD=2.6).

A locus spanning 60 cM (D10Mit61) - 65 cM (D10Mit14) on mouse Chromosome 10 showed linkage to lung inflammatory response (LOD=6.8).

A novel locus on mouse Chromosome 15 showed linkage to both lung tumor development (39.1 cM - 55.6 cM, LOD=3) and inflammatory response (46.7 cM - 67.1 cM, LOD=1.9) after treatment with urethane or urethane/butylated hydroxytoluene (BHT). This region is flanked by markers D15Mit156 (39.1 cM) and D15Mit35 (67.1 cM).

A novel locus showing linkage to urethane-induced lung tumors mapped to 43 cM on mouse Chromosome 14 (LOD=3.3 at D14Mit69).

A locus spanning 18.4 cM - 19 cM on mouse Chromosome 17 showed significant linkage to 3-methylcholanthrene (MCA)-induced tumors (LOD=7.1), and a locus spanning 18.4 cM - 31 cM showed linkage to MCA/BHT-induced tumors (LOD=3.7). These loci are thought to correspond to previously identified QTLs Pas2 (17.4 cM) and Pas12 (21.95 cM). These loci map near the H2 locus (23 cM), which is implicated in many inflammatory responses.'

02.04.2016 Curator Note: Since both Pas2 and Pas12 were originally mapped using AXB and BXA RI strains, which differ from the mapping population used here, we would not equate
either QTL with the QTL identified here on Chr 17. See J:21801 (Pas2) and J:48996 (Pas12).

A locus on mouse Chromosome 18 spanning 37 cM (D18Mit51) - 41 cM (D18Mit50) showed linkage to MCA/BHT-induced lung tumors (LOD=3.7)and a locus at 37 cM showed linkage to inflammatory response (LOD=2.1). These loci are thought to correspond to previously identified QTLs Par2 (47 cM) and Pas7 (40 cM).

02.04.2016 Curator Note: Since Par2 was originally mapped using RC mice derived from BALB/c and A/J mice (J:70565) and Pas7 was mapped using F2 and backcrosses from BALB/c and A/J mice (J:50533) we would not equate the Chr 18 QTL identified here with either of the previoulsy mapped QTL.
