Linkage analysis was performed on 26 BXD (B=C57BL/6J; D=DBA/2J) recombinant inbred (RI) strains to identify QTLs associated with cocaine-induced seizures. A panel of 655 polymorphic loci was used for the genome scan. Animals were given an IP injection of60 mg/kg cocaine solution. Parental strain C57BL/6J is more sensitive to cocaine-induced seizures compared to DBA/2J. Several loci reaching the statistical significance level of P<0.01 and P<0.05 were identified. These QTLs are deemed provisional and await further study and confirmation.

On mouse Chromosome 2, a cocaine-induced seizure locus mapped to 75 cM near Semg1 (P<0.05). DBA/2J-derived alleles confer susceptibility to cocaine-induced seizures at this locus.

On mouse Chromosome a QTL reaching P<0.05 was detected near markers D3Byu5 (33.3 cM) and D3Tu51 (43.6 cM). DBA/2J-derived alleles confer susceptibility to cocaine-induced seizures at this locus.

On mouse Chromosome 4, a QTL reaching P<0.05 was detected near D4Byu4 (66.1 cM). DBA/2J-derived alleles confer susceptibility to cocaine-induced seizures at this locus.

On mouse Chromosome 6 a QTL reaching P<0.05 mapped to 35 cM near D6Mit9, and a QTL reaching P<0.01 mapped to 60 cm on mouse Chromosome 6 near Qui (50 cM) and Cyx (50.5 cM). C57BL/6J-derived alleles confer increased susceptibility to cocaine-induced seizures at both loci.

On mouse Chromosome 7 QTLs reaching P<0.05 mapped to 13 cM near marker D7Rp2e and 57 cM near marker Rasl4. DBA/2J-derived alleles confer susceptibility to cocaine-induced seizures at D7Rp2e and C57BL/6J-derived alleles confer susceptibility to cocaine-induced seizures at Rasl4.

On mouse Chromosome 8 QTLs reaching P<0.05 mapped to 33.5 cM near D8Mit9 and 56 cM near D8Byu6. C57BL/6J-derived alleles confer susceptibility to cocaine-induced seizures at both loci.

On mouse Chromosome 12 linkage to cocaine-induced seizure susceptibility mapped to an interval spanning 56 cM - 66 cM. Markers exhibiting the strongest linkage are Crip1 and Igh at 65 cM (P<0.01) and Igh-Gte and D12Byu2 at 66 cM (P<0.01). DBA/2J-derived alleles confer susceptibility to cocaine-induced seizures at this interval.
