Advanced intercross lines (AILs) derived from A/JOlaHsd and C57BL/6JOlaHsd at the F11 generation were used to map QTLs associated with malaria parasitemia levels. AIL animals were challenged at 12- to 14-weeks of age by intraperitoneal injection with 3 x10(6) Plasmodium chabaudi parasitized red blood cells. Parental strain A/JOlaHsd is susceptible to infection by P.chabaudi whereas parental strain C57BL/6JOlaHds is resistant.

Char8 (P. chabaudi malaria resistance QTL 8) mapped to 31 cM on mouse Chromosome 11 near D11Mit242 (P<0.01). The 95% confidence interval of Char8 spans from 17 cM (D11Mit231) to 40 cM (D11Mit30). Char8 explains 2% of the phenotypic variance and appears to have an effect in the late stages of infection. Several candidate genes map to the Char8 interval, including Csf2 (29.5 cM) and the Th2 cytokines. Char8 is sytenic to a region of human Chromosome 5q31-q33 this shows linkage to Plasmodium falciparum blood infection levels in humans (pfbi).
