Linkage analysis was performed on a population of (C57BL/6 x DBA/2)F2 intercross animals to identify QTLs associated with nitrous oxide-induced antinociception. 77 polymorphic markers at an average spacing of 19 cM were used in the genome scan. Parental strain C57BL/6 exhibits greater antinociception in response to 70% nitrous oxide (NO2) compared to parental strain DBA/2.

A male-specific QTL named Noan1 (nitrous oxide antinociception 1) mapped to 41 cM on mouse Chromosome 2 near D2Mit91 (P=5x10-5). C57BL/6-derived alleles confer increased NO2-induced antinociception with partial recessive inheritance. This locus explains approximately 5.1% of the phenotypic variance.

Noan2 mapped to 89 cM on mouse Chromosome 5 near D5Mit409 (P=1.3x10-4). C57BL/6-derived alleles confer increased NO2-induced antinociception with partial dominant inheritance. This locus explains approximately 3.7% of the phenotypic variance. A candidate gene mapping near Noan2 is Nos1.

A suggestive QTL mapped to 45 cM on mouse Chromosome 18 near D18Mit186 (P=5x10-4). DBA/2-derived alleles confer increased NO2-induced antinociception with recessive inheritance. This locus explains approximately 3.1% of the phenotypic variance.

A significant interacting locus pair was detected at D6Mit116 on mouse Chromosome 6 and at D13Mit91 on mouse Chromosome 13 (P=3x10-5). High antinociception is observed when the genotype is heterozygous at D13Mit91 and homozygous C57BL/6 at D6Mit116.
