22 BXD (B=C57BL/6; D= DBA/2) recombinant inbred (RI) strains were screened for 1300 polymorphic markers to identify QTLs associated with nitrous oxide-induced antinociception. Parental strain C57BL/6 exhibits greater antinociception (sensitivity) in response to 70% nitrous oxide (NO2) compared to parental strain DBA/2 (resistant). Male animals at 5 weeks old were subjected to 70% nitrous oxide gas for this study. Several markers showed suggestive linkage to N2O sensitivity. The C57BL/6-derived allele confers increased N2O-induced antinociception at all loci.

On mouse Chromosome 2, markers Il2ra (6.4 cM) and D2Mit63 (65 cM) show linkage to N2O-induced antinociception. The region around Il2ra may be considered a probable QTL with a statistical significance of P<0.002. D2Mit63 may be considered a suggestive QTL with a statistical significance of P<0.01. The marker Gcg (29 cM) also shows suggestive linkage to N2O-induced antinociception (P<0.01). **Note: In this paper the authors place Gcg at 29 cM on mouseChromosome 6. Data in MGD places Gcg on mouse Chromosome 2.**)

On mouse Chromosome 5, marker Iapls2-5 (92 cM) shows linkage to N2O-induced antinociception. The region around Iapls2-5 may be considered a suggestive QTL with a statistical significance of P<0.01.

On mouse Chromosome 7, marker Hbb (49 cM) shows linkage to N2O-induced antinociception. The region around Hbb may be considered a probable QTL with a statistical significance of P<0.002.

On mouse Chromosome 16, marker Hmgb1-rs7 (19 cM) shows linkage to N2O-induced antinociception. The region around Hmgb1-rs7 may be considered a probable QTL with a statistical significance of P<0.002.

On mouse Chromosome 18, marker Iapls3-12 (43 cM) shows linkage to N2O-induced antinociception. The region aroundIapls3-12 may be considered a suggestive QTL with a statistical significance of P<0.01.

On mouse Chromosome 19, marker Gsl5 (12 cM) shows linkage to N2O-induced antinociception. The region around Gsl5 may be considered a probable QTL with a statistical significance of P<0.002.
