Linkage analysis was performed on 185 female animals from a (C57BL/6J x C3H/HeJ)F2 intercross. 134 polymorphic markers at an average spacing of 10 cM were used in the genome scan. Animals were place on an atherogenic diet for at least 3 weeks. Parental strain C57BL/6J is susceptible to atherosclerosis while parental strain C3H/HeJ is resistant. On an atherogenic diet, parental strain C57BL/6J exhibits decreased HDL cholesterol compared to C3H/HeJ. Parental strain C57BL/6J is also susceptible to gallstonescompared to C3H/HeJ.

Linkage to HDL cholesterol levels on both the CHOW (LOD=10.9) and atherogenic (LOD=15.2) diet mapped to distal mouse Chromosome 1 near D1Mit36 (92.3 cM). This locus is named Hdl34 (HDL level 34) and colocalizes with the Apoa2 gene (92.6 cM). Hdl34 also coincides with a previously identifided HDL QTL named Ath1 (90.1 cM). C3H/HeJ-derived alleles confer increased HDL cholesterol on both diets.

Linkage to HDL cholesterol levels (LOD=2.5) and Cyp7a1 mRNA levels (LOD=5.8) on an atherogenic diet mapped to 28 cM on mouse Chromosome 3 near D3Mit120. This locus is named Hdl35.

Linkage to HDL cholesterol levels (LOD=2.8) on an atherogenic diet mapped to 21.9 cM on mouse Chromosome 4 near D4Mit111. This locus is named Hdl36.

Linkage to VLDL/LDL cholesterol (LOD=2.6), HDL cholesterol (LOD=3.4), and Cyp7a1 mRNA levels (LOD=3.7) on an atherogenic diet mapped to 45 cM on mouse Chromosome 5 near D5Mit205. This locus is named Hdl37. Hdl37 also shows linkage to gallstone mass. The C3H/HeJ-derivedallele is associated with decreased gallstone formation at Hdl37. A potential candidate gene is Tcf1 (65 cM).

Linkage to total cholesterol (LOD=3) and HDL cholesterol (LOD=4) on the CHOW diet mapped to 16 cM on mouse Chromosome 7 near D7Mit227. This locus is named Hdl38. Potential candidate genes near Hdl38 are Lipe (5.5 cM), Saa (23.5 cM), and Apoe (4 cM). Suggestive linkage to Cyp7a1 mRNA levels on an atherogenic diet mapped to 72.4 cM near D7Nds4 (LOD=2.9).

Linkage to HDL cholesterol (LOD=2.5) and Cyp7a1 mRNA levels (LOD=4.4) on an atherogenic diet mapped to 37 cM on mouse Chromosome 11 near D11Mit4. This locus is named Hdl39. Animal heterozygous at Hdl39 exhibit increased HDL cholesterol levels on an atherogenic diet. Hdl39 also shows linkage to gallstone formation. The C3H/HeJ-derived allele is associated with decreased gallstone formation at Hdl39. A potential candidate gene is Tcf2 (44 cM).

Linkage to Cyp7a1 mRNA levels (LOD=3.9) on an atherogenic diet mapped to 58 cM on mouse Chromosome 12 nearD12Mit8. This locus is named C7mlq (Cyp7a1 mRNA level QTL).

Linkage to total cholesterol (LOD=4.4) and HDL cholesterol (LOD=3.5) on the CHOW diet, and HDL cholesterol on the atherogenic diet (LOD=3.0) mapped to 32 cM on mouse Chromosome 18 near D18Mit124. This locus is named Hdl40.
