Linkage analysis was performed on 196 (C57BL/6J x MSM/Ms)F1 x MSM/Ms backcross animals to identify QTLs influencing sensitivity to ether anesthesia. Parental strain C57BL/6J exhibits resistance to ether anesthesia whereas parental strain MSM/Ms is sensitive. (C57BL/6J x MSM/Ms)F1 hybrids are resistant to ether anesthesia. An initial screen was done with 40 microsatellite markers using 12 resistant and 12 sensitive backcross animals. Linkage was detected on mouse Chromosomes 3, 7, and 8. More markers weretyped in the remaining animals and mouse Chromosome 7 showed the strongest linkage between 23 cM (D7Mit313) and 29 cM (D7Nds1). Peak linkage occurred near D7Nds1 with LOD=3.03. This locus is named Etan1 (ether anesthesia 1). Animals heterozygous at Etan1 are resistant to ether anesthesia compared animals homozygous for MSM/Ms-derived alleles. Potential candidate genes mapping near Etan1 are Gabrg3 (28.2 cM), Gabra5 (28.5 cM), Gabrb2 (28.6 cM), and Chrna7 (30 cM).
