The QTL interval of Alcp1 (alcohol preference locus 1, male specific) on mouse Chromosome 2 was further refined in this study using interval specific congenic recombinant strains (ISCRSs). ISCRSs were constructed using inbred parental strains DBA/2J (donor) and C57BL/6J (background). Several lines with various DNA breakpoints between D2Mit365 (17 cM) and D2Mit351 (47.5 cM) were constructed. Parental strain C57BL/6J exhibits greater alcohol preference compared to parental strain DBA/2J.

Three ISCRSs were identified which exhibited the low alcohol preference phenotype of the DBA/2J donor parent. Comparison of the DBA/2J-derived regions of these ISCRSs suggests that Alcp1 may be composed of two separate putative loci from D2Mit56 (41 cM) to D2Mit10 (44 cM), and from D2Mit10 (44 cM) to D2Mit351 (47.5 cM). Further study is required to confirm if Alcp1 is composed of 2 tightly linked QTLs.
