    Linkage analysis was performed on 62 animals from a (C57BL/6J x CBA/J)F2 intercross to identify QTLs associated with hepatic immunopathology traits induced by the parasite Schistosoma mansoni. Animals were infected with S. mansoni at 6 weeks of age and studied 8 weeks post-infection. Parental strain CBA/J infected with S. mansoni exhibits larger granulomas and increased IFN-gamma, IL-2, IL-5, and IL-10 production compared to parental strain C57BL/6J. 82 markers spanning the 19 autosomes with an average spacing of 15.5 cM were used for the genome scan.

    Suggestive linkage to granuloma size mapped to mouse Chromosome 13 near D13Mit3 (10 cM; LOD=2.2). The CBA/J-derived allele confers increased granuloma size with a recessive mode of inheritance. Potential candidate genes for this locus are Serpinb1c (12.2 cM), Serpinb6b (12.3 cM), and Serpinb9 (12.4 cM).

    Suggestive linkage to IFN-gamma production mapped to mouse Chromosome 1 near D1Mit151 (101 cM; LOD=2.6). The CBA/J-derived allele confers increased IFN-gamma production with a dominant mode of inheritance. Potential candidate genes for this locus are Parp1 (98.6 cM) and Tlr5 (98 cM). This locus is syntenic to a region on human Chromosome 1 that shows linkage to S. mansoni infection levels in humans.

    Suggestive linkage to IFN-gamma production mapped to mouse Chromosome 5 near D5Mit81 (28 cM; LOD=3.2). The CBA/J-derived allele confers increased IFN-gamma production with a recessive mode of inheritance.

    The IFN-gamma loci at D1Mit151 and D5Mit81 show interaction. Animals with a CBA/J homozygous or heterozygous genotype at D1Mit151 and a CBA/J homozygous genotype at D5Mit81 exhibit the highest levels of IFN-gamma production. Animals with this genotype also exhibit increased granuloma size.
