    Linkage analysis was performed on 102 male (C3H/Or x NOD)F1 x NOD backcross animals to identify QTLs associated with streptozotocin (STZ)-induced diabetes. STZ is a drug that when administered at high doses destroys beta pancreatic cells and results in diabetes in NOD mice but not in C3H/Or mice. F1 hybrid animals appear resistant to STZ-induced diabetes in a similar fashion to the C3H/Or parental. 74 autosomal microsatellite markers at an average spacing of 20 cM - 30 cM were used for the genome scan.

    A locus on mouse Chromosome 11 near D11Mit286 (52 cM) shows linkage to STZ-induced diabetes (P=0.07). This locus is named Stzid (streptozotocin induced diabetes susceptibility). The NOD-derived allele at Stzid confers susceptibility to STZ-induced diabetes and appears to be inherited recessively. Nos2 at 45.6 cM is a potential candidate gene for Stzid because Nos2 inactivation has been shown to prevent multiple low dose streptozotocin induced diabetes in a different cross. Nos2 knockout animals on a C57BL/6 STZ-susceptible background were studied. Knockout animals were sensitive to STZ-induced diabetes similar to background strain C57BL/6. Therefore, Nos2 does not appear to be responsible for the Stzid phenotype. Stzid colocalizes with the insulin dependent diabetes susceptibility QTL Idd4 (46 cM).

    A locus on mouse Chromosome 9 near D9Mit135 (48 cM) shows linkage to STZ-induced diabetes (P=0.036). This locus is named Stzid2 (streptozotocin induced diabetes susceptibility 2). The NOD-derived allele at Stzid2 confers susceptibility to STZ-induced diabetes and appears to be inherited recessively.

    Animals homozygous for NOD-derived alleles at both Stzid and Stzid2 exhibit significantly increased susceptibility to STZ-induced diabetes.
