A locus on mouse Chromosome 3 shows association with longer circadian rhythm in a congenic strain carrying the Car2 null mutation. The B6.D2-Car2n/J congenic carries a DBA/2J-derived Car2n region of mouse Chromosome 3 between D3Mit60 (0 cM) and D3Mit221 (2.4 cM) on a C57BL/6J genetic background. Two different recombinant inbred strains named R1 and R2 were created from B6.D2-Car2n/J. The recombinant congenic R1 carries DBA/2J-derived DNA from D3Mit164 (2.4 cM) to D3Mit221 (2.4 cM) on a C57BL/6J genetic background. The recombinant congenic R2 carries DBA/2J-derived DNA from D3Mit60 (0 cM) to D3Mit149 (2.4) on a C57BL/6J genetic background.

The R1 line exhibits increased circadian rhythm compared to the R2 line and C57BL/6J control animals. This finding suggests that a QTL for circadian rhythm is localized to a 1 cM interval on mouse Chromosome 3 encompassing D3Mit164 and D3Mit221. This locus is named Cplaq8 (circadian period of locomotor activity 8). An allele assay showed that Car2 is located within this interval and it is considered a candidate gene for Cplaq8.
