A previously identified QTL on mouse Chromosome 4 named Thyls (thymic lymphoma susceptibility) is linked to susceptibility to gamma-irradiation induced thymic lymphomas. Thyls shows linkage to D4Mit12 at 57.6 cM. In this study, association of Thyls to MNU-induced thymic lymphomas was investigated. A congenic strain carrying the Thyls locus was previously derived from MSM donor and BALB/c recipient strains. The congenic region spans a 25 cM interval flanked by D4Mit9 (44.5 cM) and D4Mit203 (60 cM). Background strain BALB/c is susceptible to gamma-irradiation induced thymic lymphomas whereas donor strain MSM is resistant.

Congenic animals were crossed to BALB/c and then to MSM. 91 backcross animals were injected with MNU and followed for 250 days. Suggestive linkage to MNU-induced thymic lymphoma susceptibility was detected at markers D4Mit12 and D4Mit338. This region coincides with the Thyls locus and suggests that Thyls or a closely linked locus is responsible for gamma-irradiation and MNU-induced thymic lymphomas. The BALB/c-derived allele at Thyls confers susceptibility to MNU-induced thymic lymphomas. A previously identified MNU-induced lymphoma QTL named Tlag2 at 53 cM maps near Thyls. A potential candidate gene for Thyls is Mycl1 at 55 cM. Thyls is syntenic to a region on human Chromosome 1p34-p36.
