Quantitative trait loci (QTL) analysis was used to identify three genes that influence tolerance and/or sensitivity to the hypothermic effect of alcohol. 907 female animals from 19 BXD (B=C57BL/6J; D=DBA/2J) recombinant inbred (RI) strains were analyzed at over 800 polymorphic loci across the genome. Further work is planned by the authors to precisely map these loci. It is believed that up to 50% of the loci mapped in this study may be false-positive.

Htas1 (hypothermia due to alcohol sensitivity 1) mapped to 83.6 cM on mouse Chromosome 1 near Prdx6 (formerly Ltw4). This locus is associated to alcohol-induced hypothermia at a dose of 3 g/kg. The Htas1 locus appears to have a pleiotropic effect as determined in other studies, with influences on amphetamine hyperthermia, alcohol acceptance drinking, and nitrous oxide withdrawal severity.

Htas2 (hypothermia due to alcohol sensitivity 2) mapped to 59 cM - 60 cM on mouse Chromosome 4 near Ahd1 (66.1 cM). C57BL/6J-derived alleles at Htas2 confer increased sensitivity to alcohol-induced hypothermia at doses of 2 g/kg and 3 g/kg. Htas1 also maps near Akp2 (70.2 cM).

Htas3 (hypothermia due to alcohol sensitivity 3) mapped to 5 cM on mouse Chromosome 5 near D5Mit1. DBA/2J-derived alleles at Htas3 confer increased sensitivity to alcohol-induced hypothermia at the 3 g/kg dose. Htas3 also maps near Drd5 (23 cM).

Htas4 (hypothermia due to alcohol sensitivity 4) mapped to 21 cM on mouse Chromosome 16 near D16Mit3. C57BL/6J-derived alleles at Htas4 confer increasedsensitivity to alcohol-induced hypothermia and alcohol tolerance at the 4 g/kg dose. Htas4 also maps near Drd3 (23.3 cM).

Htas5 (hypothermia due to alcohol sensitivity 5) maps to 62 cM - 65 cM on mouse Chromosome 13 near Rasa1. This locus is associated with sensitivity to alcohol-induced hypothermia at the 2 g/kg dose. Htas5 also maps near Rasl4 (55 cM) and Arsb (formerly As1; 50 cM).

Htas6 (hypothermia due to alcohol sensitivity 6) mapped to 60 cM on mouse Chromosome 6 near Qui. This locus is associated with sensitivity to alcohol-induced hypothermia at the 2 g/kg dose.

Htas7 (hypothermia due to alcohol sensitivity 7) mapped to 56 cM on mouse Chromosome 9 near Gnat1. This locus is associated with sensitivity to alcohol-induced hypothermia at the 4 g/kg dose.

Htas8 (hypothermia due to alcohol sensitivity 8) mapped to 9 cM on mouse Chromosome 19 near Pomc-ps1. This locus is associated with alcohol tolerance at the 4 g/kg dose.

Htas9 (hypothermia due to alcohol sensitivity 9) mapped to 30 cM - 50 cM on mouse Chromosome 2. This locus has been implicated in several mapping studies involving alcohol sensitivity. Potential candidate genes mapping near Htas9 are Gad1 (43 cM), Scn1a (36 cM), Scn2a1 (36 cM), and Scn3a (36 cM).
